Changes of Expression of Synaptophysin(SYN)in the Different Parts of Rats’ Central Nervous System Following Spinal Cord Injury

摘要 目的研究脊髓损伤后突触素(SYN)在中枢神经系统中不同部位的变化,为脊髓损伤研究提供实验依据.方法成年健康SD大鼠42只,随机分成7组(假损伤组;1、3、7、14、21和28d脊髓挫伤组).观察各组SYN在大鼠脊髓、大脑、脑干、海马的表达分布情况.用图像分析系统测SYN阳性单位和平均光密度.结果脊髓损伤后,脊髓和延髓段的SYN阳性单位值均在1d组即开始下降,14d到最低值,与对照组差异有统计学意义(P<0.05),以后逐渐增高;海马区SYN阳性单位值则在3d达最低值,与对照组有明显差异(P<0.05);其它部位SYN表达为阴性.SYN阳性单位值的变化趋势与其平均光密度值变化类似.结论脊髓损伤后,在中枢神经不同部位中的突触素表达有不同的变化规律. Objective To provide the experimental evidences for the studying of mechanisms of the spinal cord injury by studying the changes of synaptophysin (SYN) in the different regions of the central nerve system. Method 42 adult SD rats were divided into 7 groups randomly (sham injury group; 1, 3, 7, 14, 21, 28 day contused spinal cord injury group). We observed the distribution of SYN in the spinal cord, brain, brain stem, cornu ammonis of SD rats. The positive unit and the average optical density were measured by image analysis system. All data were analyzed via statistical software SPSS. Results Positive unit of SYN in the spinal cord and oblongata began to fall on the 1st day group after spinal cord injury, then reached the nadir on the 14th day and there was significant difference between the experimental group and control group (P 〈 0.05) , and increased gradually. Positive unit of SYN in hippocampus reached the nadir on the 3rd day and there was significant difference between the experimental group and the control group (P 〈 0.05) , the rest were negative. The changes of positive unit in SYN were similar to the average optical density. Gonclusions After spinal cord injury, the changes of expressions of the synaptophysin (SYN) in the different regions of the central nerve system is different.
