摘要
目的:通过观察肾脏组织形态学及肾小球α-SMA表达的变化,探讨祛风通络方防治大鼠系膜增生性肾小球肾炎的可能机制,为祛风通络方的临床应用提供理论依据.方法:采用免疫法制备MsPGN大鼠模型,光镜下观察各组大鼠肾小球系膜区(GMC)及细胞外基质(ECM)积聚变化.采用免疫组织化学法检测各组大鼠肾组织中α-SMA的表达,并用图像分析系统对免疫组织化学结果进行灰度值分析.结果:肾小球及肾小管α-SMA灰度值病理组为167.91±6.16,空白对照组为195.88±3.49,两者相比较差异具有统计学意义(P<0.05);肾小球及肾小管α-SMA灰度值祛风通络方组为200.71±2.29,蒙诺组为201.68±1.37,与病理组相比,2组灰度值明显升高(P<0.05),2治疗组与空白对照组相比灰度值虽有所升高但差异无显著性.肾组织形态学观察显示,祛风通络方组个别区域肾小球系膜细胞轻度增生,系膜区轻度增宽,管腔无挤压现象,较模型组明显改善.结论:祛风通络方能够抑制肾组织α-SMA的表达,影响肾小球系膜细胞的重塑,该方可减轻MsPGN模型大鼠肾组织病理损害,延缓肾小球硬化的进展.
AIM: To study the effect and mechanism of Qufeng- tongluo(QFTL) Recipe in treating rats with mesangial proliferative glomerulonephritis (MsPGN) so as to provide some theoretic basis for the clinical application of Qufengtongluo Recipe. METHODS: A total of 48 rats were randomly divided into 4 groups(n = 12): Blank, model and two treatment groups(QFTL and Monopril, respectively ). MsPGN models were made by immunization in the 3 groups except in blank group. Mesangial ceils(GMC) and extracelluar matrix(ECM) were observed with light microscope in each group. The expression of α-SMA was observed with the immunohistochemical staining and the grey of immunohistochemical staining was analyzed with the image analysis system. RESULTS: The grey of α-SMA in glomerulus and tubules significantly decreased ( P 〈 0. 05 ) in the model group. In the 2 treatment groups, the grey of α-SMA was enhanced significantly compared with that in model group (P 〈 0.05 ), while no statistics significance was observed compared with that in blank group. No statistics significance was observed between the 2 treatment groups. In the QFTL group, the morphological changes in renal tissues showed that mesangial cells proliferated lightly and the regions of mesangial matrix broadened lightly, with no extruded phenomenon in renal tubule cave. These results were much better than those in model group. CONCLUSION: QFTL Recipe refrains the expression of α-SMA and influences the remolding of GMC. This recipe reduces pathological lesions of nephridial tissues in MsPGN rats and delays the process of glomerular sclerosis.
