摘要
目的研究电脉冲对不同卵龄小鼠MⅡ期卵母细胞电激活影响,寻找小鼠体细胞克隆避免电激活的适宜卵龄的卵母细胞。方法在180V/mm,9s,1个电脉冲(1p)下,比较了注射人绒毛膜促性腺激素(hCG)后13,16h卵母细胞经体外培养(IVC)4h的激活率、碎裂+死亡率。结果hCG后13h和16h的MⅡ期卵母细胞相比,激活率分别是6.2%和29.5%,两者间差异有极显著性意义(P<0.01)。碎裂+死亡率两组间没有显著性差异。结论注射hCG后13h的卵母细胞为适宜的小鼠核移植的受体细胞,在一定程度上可以有效的避免电激活重构胚。
Objective To research the influence of electric pulse for different age of mouse egg, to avoid electrical activation of the appropriate age, to find eggs oocytes mouse somatic cloning cells. Methods The rate of the activated egg and the rate of lysed+dead egg were compared at different ages of oocytes after in vitro cultured 4h: 13, 16h (after HCG injection) with the electrical parameters of 180V/mm 9 s, 1 electrical pulses (lp). Results The activation rate of 13h and 16h ooeytes was high up to 6.2% and 29.5 % respectively, the difference between them was significant (P (0.01). But it was not different about the lysed +dead rate in the two groups. Conclusions After injection HCG 13 h,oocytes which are suitable for the receptor cell in mice nuclear transfer, can effectively avoid activated reconstruction embryo.
