一贯煎抗无水乙醇性胃粘膜损伤的作用

Studies on effects of yiguanjian against gastric mucosal damage induced by absolute ethanol *

目的研究一贯煎对实验性胃粘膜损伤的影响，并分析其作用的主要成分和机制．方法给大鼠胃饲不同剂量一贯煎煎液，观察对无水乙醇所致胃粘膜损伤的影响．另用一贯煎去生地煎剂胃饲和预先肌注消炎痛等方法进行分析．结果胃饲一贯煎６，１２和２４ｇ／ｋｇ，随着剂量增加，显著减轻无水乙醇性胃粘膜损伤，损伤指数依次为１００７±１４７，６６１±１３３和６７±３８，损伤抑制率依次为６７％，３７４％和９３４％．一贯煎去生地煎剂２４ｇ／ｋｇ对胃粘膜损伤无明显影响．消炎痛５ｍｇ／ｋｇ不能取消一贯煎抑制胃粘膜损伤作用．结论一贯煎能有效防止无水乙醇对胃粘膜的损伤，其作用与主药生地密切相关，而与前列腺素（ＰＧｓ）的合成和释放无关．

AIM To study the effects of Yiguanjian on experimental gastric mucosal damage and analyze its active components and mechanisms of action. METHODS Different doses of decoction of yiguanjian were administered orally to rats to observe the inhibition of gastric mucosal damage induced by absolute ethanol. The dry Radix Rehmannia was removed from Yiguanjian and indomethacin was injected intramuscularly in advance for comparative analysis. RESULTS The decoctions of Yiguanjian administered orally at doses of 6, 12 and 24g/kg inhibited remarkably gastric mucosal damage in a dose dependent manner. The injury index was 100 7±14 7,66 1±13 3 and 6 7±3 8; and the damage inhibitory rate was 6 7%, 37 4% and 93 4% respectively. Yiguanjian excluding the dry Radix Rehmannia could not prevent damage at a dose of 24g/kg . Indomethacin 5mg/kg could not cancel the inhibitory effect of Yiguanjian on gastric mucosal damage. CONCLUSION Yiguanjian can prevent ethanol induced gastric mucosal damage, which may be produced by the dry Radix Rehmannia, a main component of yiguanjian and not mediated by prostaglandin.
