摘要 目的比较紫杉醇耐药卵巢癌细胞SKOV3/TAX及敏感细胞SKOV3基因表达谱的差异,筛查获得性紫杉醇卵巢癌耐药的相关基因。方法取对数生长期SKOV3、SKOV3/TAX细胞,抽提细胞总RNA,合成双链cDNA,生物素标记,用高通量Affymetrix人类基因组U133A基因芯片检测其基因谱。结果与SKOV3相比,SKOV3/TAX有111条基因表达发生变化(45条基因表达上调,66条基因表达下调)。结论SKOV3/TAX的部分基因表达发生变化,与卵巢癌化疗耐药有关。 Objective To investigate tile gene differential expression of taxol-resistant ovarian cancer SKOV3/TAX cell line compare with its parental cell SKOV3, and to screen the candidate gene with taxol-resistance. Methods The total RNA from tile SKOV3/TAX and its parental cell SKOV3 were extracted respectively during the log phase. Then double strand cDNA was compounded. The differential were detected by high flux of Affymetrix human U133A cDNA gene chips. Results Total 111 differential expression genes were screen out in SKOV3/TAX cell compared with SKOV3 cell, inclu- ding 45 up - regulated genes and 66 down - regulated genes. Conclusion These differential expression genes in SKOV3/ TAX cells are related with the mechanism of drog-resistance that involves in ovarian cancer chemotherapy.
