摘要 目的 探讨突变型K—ras蛋白在非小细胞肺癌(NSCLC)组织中的表达及临床意义。方法应用免疫组化S—P法,检测56例非小细胞肺癌(NSCLC)、45例正常肺组织(NL)中K—ras蛋白的表达。结果K—ras蛋白总阳性率为32.14%(18/56),鳞癌、腺癌、鳞腺癌、大细胞癌阳性率分别为26.67%(8/30)、53.33%(8/15)、16.67%(1/6)、20.00%(1/5),K—ras蛋白在肺正常组织为阴性。在鳞癌、腺癌K—ras均主要表达于胞浆,K—ras的阳性表达与肺癌组织学类型、肺癌分期、吸烟及有无淋巴结转移等有关,与肿瘤分化程度无关。结论K—ras基因的突变参与了非小细胞肺癌的发生和发展,检测肺癌K—ras蛋白表达可作为判断其预后的指标。 Objective To investigate the clinical significance and the expression of mutated K - ras in non - small cell lung cancer (NSCLC). Methods Immunohistochemical method was used to examine K - ras protein expression of the sections from 56 cases of NSCLC and 45 normal lung tissues (NL). Results The positive rate of K - ras expression in squamous cell carcinoma, adenocarcinoma, adenosquamous carcinoma and large cell carcinoma was 26.67% (8/30) , 53.33 % ( 8/15 ) , 16.67 % ( 1/6 ) , 20.00% ( 1/5 ) respectively. The positive rate of adenocarcinoma was significantly higher than that of normal lung tissue and squamous cell carcinoma( P 〈 0.05 ). Besides, expression area of K - ras was often in the eytoplasma for squamous cell carcinoma and adenocarcinoma. The expression of K - ras gene was not related to differentiation, but the expression of K -ras gene was related to pathological type, smoking, stage and lymph nodes metastasis. Conclusions The results suggest that overexpression of K - ras involve in genesis and development of non - small cell lung cancer (NSCLC). Detection of K - ras protein can be used as a prognostic factor for the patients.
