摘要
目的研究顺铂对人肺癌LTEP-a-2细胞DNA损伤相关基因表达的影响。方法应用CCK-8法检测顺铂对LTEP-a-2细胞增殖的影响,流式细胞术检测细胞周期和凋亡的改变,PCR检测DNA损伤相关基因表达的改变。结果顺铂对LTEP-a-2细胞的增殖呈抑制作用,细胞周期发生改变,细胞被阻滞于G2/M期,并诱导凋亡,PCBP4、TP73基因表达上调,DDB1、LIG1基因表达下调。结论顺铂对LTEP-a-2细胞的DNA损伤相关基因表达有影响,其作用与DNA损伤有关。
Objective To observe the effects of cisplatin on the expression of genes involved in DNA damage response in human lung cancer cell line LTEP-a-2. Methods The effect of cisplatin on the growth of hunman lung caner cell line LTEP-a-2 was studied with CCK-8 assay, that on the cell cycle phase distribution and apoptosis was determined using flow cytometry and that on the expression of genes involved in DNA damage response in human lung cancer cell line LTEP-a-2 was studied with PCR assay. Results Cisplatin inhibited the proliferation of LTEP-a-2 cells. LTEP-a-2 cells treated with cisplatin were accumulated in G2/M phase in the cell cycle, and apoptosis was induced. The expressions of PCBP4 and TP73 were increased, while DDB1 and LIG1 were decreased. Conclusion The expressions of genes involved in DNA damage response are changed in LTEP-a-2 cells treated with cisplatin. The action of cisplatin may be related to DNA damage.
