外源性TNF-α对bBMP异位诱导成骨的影响 被引量:2

The Effects of Recombinant Human Tumor Necrosis Factorα on Heterotopic Bone Formation Induced by Bovine Bone Morphogenetic Protein

摘要 目的:了解局部注射重组人α肿瘤坏死因子(rhTNF-α)对牛骨形态发生蛋白(bBMP)在小鼠体内异位诱导成骨的影响。方法:采用X线检查成骨面积图像分析、碱性磷酸酶活性测定、钙含量测定和组织学检查等方法,对局部注射不同剂量rhTNF-α后bBMP诱导成骨活性的变化进行评价。结果:①bBMP5mg植入小鼠肌袋内可诱发一个完整的骨发育过程,导致异位骨软骨组织发生;②在bBMP植入区注射rhTNF-α后,bBMP诱导成骨活性受到干扰,表现为成骨活性随rhTNF-α注射剂量的增加而减小;③rhTNF-α使bBMP诱导的间充质细胞分化成骨过程受到影响,导致成骨时间延长或完全阻滞。 Objective: In investigate the effects of local injection of recombinant human tumor necrosis factorα(rhTNFα) on heterotopic bone formation (HBF) induced by bovine bone morphogenetic protein (bBMP) in the muscle porch of mice. Methods: In mice, bBMPinduced HBF receiving injections of different doses of rhTNFα were assessed quantitatively by computer xray image analysis, alkaline phosphatase activity, calcium content, and histological examination. Results:1. A natural process of bone formation was induced by 5mg of bBMP. 2. The osteogenic ability of bBMP was interfered with injections of rhTNFα. The more rhTNFα injected, the less osteogenesis induced. 3. Local injections of rhTNFα inhibited the transformation of the mesenchymal cells into osteogenic cells and the bone development. The osteogenisis induced by bBMP was delayed or stopped. Conclusion: This study suggests that rhTNFα can reduce the bBMPinduced HBF and inhibit the process of bone development.
