小鼠胸腺基质细胞单抗体特性及生物功能研究

摘要 应用细胞ELISA用间接荧光染色技术,分析多种胸腺基质细胞(TSC)单克隆抗体,结果表明不同的单抗与不同TSC呈不同的阳性反应,其中Pf18-3和Rs21-C6单抗与小鼠胸腺基质细胞1-7(MTSC1-7)等多种细胞均呈强阳性反应,但荧光染色显示Pf18-3和Rs21-C6染色特征不一。将Pf18-3或Rs21-C6单抗加入TSC与胸腺细胞共培养,发现两种单克隆抗体对胸腺基质细胞和胸腺细胞结合有明显抑制作用:Pf18-3、Rs21-C6对MTSC1与胸腺细胞结合的抑制率分别为64.3%、41.3%;对MTSC4与胸腺细胞结合的抑制率分别为60.4%和38.8%。试验表明Pf18-3、Rs21-C6两种单抗分别识别胸腺基质细胞表面抗原分子,且其识别的抗原分子参与了胸腺基质细胞与胸腺细胞的结合。 eries of monoclonal antibodies(mAbs)specific for MTSC(mouse thymic stromal cells)were observed by cell ELISA and indirect immunofluorescence staining.Results indicated that defferent mAb showed different reaction to various MTSC lines.Among of them mAbs,Pf18-3 and Rs21-C6 showed strong positive reation to MTSCs.Immunofluorescence staining found that the staining charactrization of two mAbs were different.The function of two mAbs were investigated further by the addition of them to the co-culture of MTSC and thymocytes.Two Ambs inhibited obviously the effect of thymocytes binding to MTSC,the inhibitory rates of Pf18-3 and Rs21-C6 were 64.3%,41.3% for for MTSCl and 60.4%,38.8% for MTSC4.There results suggested that the molecules detected by Pf18-3 and Rs21-C6 may mediate the binding between MTSC and thymocytes.
