摘要
目的实验研究神经肽Y测定在诊断心脑血管及胃肠系统疾病的生理病理学意义。方法以神经肽Y甲状腺球蛋白联接物为免疫原,注射家兔获得高质量抗体,建立了神经肽YRIA方法,并组装试剂盒。结果本法回收率平均为98%,批内批间CV分别为<65%和<110%,最小检出值<33ng/L,反应最佳条件为005mol/LpH8.6巴比妥缓冲液。测定28例正常人血浆NPY为1451±441pg/ml。结论神经肽YRIA灵敏度高、特异性强,在生物医学实验研究中是一项有实用价值的指标。
Objective To develop a RIA kits for measurement of neuropeptide Y(NPY).Methods Chemically conjugated NPYTG used as antigen was injected into the rabbit for immunization. The highly qualitative antibody against NPY was prepared from immunized rabbit serum. NPY was labelled with 125I. A NPY RIA was established and the NPY RIA kits were made up. Results The average recovery rate of NPY was 98%. The intra and interassay CVs were <6.5% and 11.0% respectively. The kits could detect a lower limit of <33 pg/ml. The best reactive condition was under pH8.6, 0.05 mol/L barbital buffer. The plasma levels of NPY in health adults were 145.1±44.1 pg/ml (±s).Conclusion The NPY RIA was a highly sensitive and specific assay. It may serve as a new applicable parameter in the biomedical experiment.
