摘要
目的观察鸡脊髓及背根神经节(DRG)中钙离子通道α2(CCα2)亚基的免疫学表达,并观察在DRG中CCα2与calbindin-D28k(CB)的共表达的相互关系。方法通过免疫组织化学,观察了鸡背根神经节中表达情况,利用图像分析系统对VDCC-α2δ-1亚单位免疫阳性和阴性神经元数量和细胞面积进行测量,统计出不同大小细胞的构成比。结果在脊髓背角I、II层和背内侧束(LT)内,CCα2δ亚基免疫阳性呈点状样分布于神经终末之中。在DRG神经元中,约72%的神经细胞呈CCα2免疫阳性,其中小型至中型神经元占免疫阳性神经元的88%。约75%的CCα2免疫阳性神经元与CB共存,其中大部分(90%)神经元是小、中型神经元。结论CCα2主要表达于伤害性神经元中,并在DRG伤害性感觉神经元中,伤害性信息的传递可能与细胞的Ca2+调节机制,即通过CCα2δ亚基的Ca2+内流和细胞内CB的Ca2+结合活性这两种因素有关。
Objective To study the distribution of the calcium channel α2 (CCα2)subunit in the chicken spinal cord and dorsal root ganglion(DRG)neurons and determine its relationship with calbindin-D28k (CB) in the DRG neurons. Methods Used immunohistochemieal methods, the number and size of immunoreactive(IR) neurons were measured by using a image analyze system, and were compared with those of negative neurons. Results CCα2 subunit was detected in nerve terminals, which were observed as dot-like structures, and in laminae I, II,III and Lissauer' s tract in the spinal dorsal horn. In the DRG neurons,approximately 72% of the total neurons were CCα2 subunit positive,and most(88% )of these neurons were small to medium sized. A majority(75% )of the positive neurons showed CB immunoreaetivity and most(90% )of these neurons were small to medium sized. Conclusion It is postulated that the mode of nociceptive transmission may involve a cellular Ca^2+ -regulating system that consists of both Ca^2+ entry via CCs with the α2 subunit and intracellular Ca^2+-binding activity of CB in the nociceptive neurons of the DRG.
