套式PCR技术用于疟疾诊断的研究

摘要 采用三对引物建立了套式PCR疟疾诊断技术,对来源于同一地区的34例门诊镜检确诊为间日疟的血标本及国外输入的2例恶性疟的血标本进行了病原体检测和虫种鉴定,得到预期扩增的DNA片段,经琼脂糖凝胶电泳,显示出肉眼可见的与引物对应的扩增区带,与镜检结果一致。 Nested polymerase chain reaction has been applied for detecting rnalaria by using 3 primers synthesised with reference to the SSU rRNA gene sequence of human Plasmdium. Blood samples from 34 proved vivaxmalaria cases in Mingshan County, Sichuan and 2 imported falciparum malaria cases were collected for Plasmodum detection and species identification. The results showed that a 120 bp amplified DNA fragment of P.vwax and 205 bp of P. falciparum were obtained and relevant bands could be seen by naked eye in argarosegel electrophoresis.
