VHL基因和HIF-1α在大肠癌组织中的表达及临床意义

The Expression of VHL Gene and HIF-1α Protein in Colorectal Carcinoma Tissues and their Clinical Significance

摘要 为探讨VHL基因和缺氧诱导因子-1α(HIF-1α)在大肠癌中的表达及其与大肠癌临床病理因素的关系,我们用免疫组织化学SP法检测165例大肠癌和20例癌旁组织中VHL、HIF-1α表达情况,并分析其表达与大肠癌中医证型、分化程度、临床分期、转移及预后的相关性。结果显示,VHL基因在165例大肠癌组织中的阳性表达率为21.8%,在20例癌旁组织中的阳性表达率为75.1%(P〈0.001);HIF-1α蛋白在165例大肠癌组织中的阳性表达率为75.2%,在20例癌旁组织中的阳性表达率为5.0%(P〈0.001);VHL基因的阴性表达与HIF-1α蛋白的阳性表达与大肠癌患者的中医证型、肿瘤分期、淋巴结转移、器官转移及预后有关(P〈0.05)。结果表明,VHL基因表达下调和HIF-1α蛋白表达上调在大肠癌的侵袭、转移中发挥重要作用。 The expression of yon Hippel-Lindau (VHL) gene and hypoxia-inducible factor-1α(HIF-1α) protein in colorectal carcinoma was measured and the correlation between the expression and the clinicopathological factors of colorectal carcinoma was evaluated in the study. The expression of VHL gene and HIF-1α protein of colorectal carcinoma in 165 cases and that of pericarcinomatous tissues in 20 cases was detected with immunohistochemistry assay,and its correlation with symptoms and signs of TCM,differen- tiation, clinical stage, metastases and prognosis of colorectal carcinoma was analized. As results, the positive expression rate of VHL gene in carcinomatous tissues of 165 cases and in pericarcinomatous tissues of 20 cases was 21.8% and 75.0%( P 〈0. 001) respectively;while the positive expression rate of HIF-1α protein in the carcinomatous tissues and in the pericarcinomatous tissues was 75.2% and 5.0% ( P〈0. 001) respectively;the negative expression of VHL gene and the positive expression of HIF-1α protein had correlation with the symptoms and signs of TCM, stage,lymph node metastasis(or metastasis of organs) and prognosis( P〈0.05). It is concluded that the down-regulated expression of VHL gene and the up-regulated expression of HIF-1α protein play an important role in the invasion and metastases of colorectal carcinoma.
