摘要
研究了有机介质中酶促选择性合成阿糖胞苷氨基酸衍生物.实验研究合成了4种L-氨基酸乙烯酯为酰化试剂;以枯草杆菌碱性蛋白酶为催化剂,在50℃条件下,L-氨基酸乙烯酯与阿糖胞苷反应合成系列阿糖胞苷5′-O-L-氨基酸酰化衍生物;产物用FTIR、NMR和ESI-MS表征确定其结构;发现不同的酰化试剂对反应产率有影响,其中以L-苯丙氨酸乙烯酯作为酰化试剂时,反应产率最高,达到了82.9%.该方法具有条件温和、选择性高和产率好等特点.
A regioselective enzymatic synthesis approach was developed in organic media for the preparation of cytarabine aminoacyl derivatives. Four different L-amino acid vinyl esters were synthesized. Then transesterification of cytarabine using L-amino acid vinyl esters as acylating reagents were performed under the catalysis of Subtilisin at 50℃, which displayed high regioselectivity. A series of 5'-O-L-aminoacyl-cytarabines were prepared with high yields and the structures of products were characterized by FTIR, NMR and ESI-MS, The influence of the acylating reagents was investigated. The highest yield was 82. 9% when L-phenylalanyl vinyl ester was used as acylating donor.
