Survivin与p21waf-1在大鼠肝细胞癌变过程中的动态表达及其相关性的研究 被引量:1

摘要 目的:动态观察黄曲霉毒素B1(AFB1)在诱发大鼠肝细胞癌(HCC)过程中survivin及p21waf-1表达的变化及它们在HCC发生发展中的关系。方法:用免疫组化及RT-PCR检测24只用AFB1诱发出的大鼠肝癌及癌旁肝组织、6只未诱发出肝癌的大鼠及11只对照大鼠不同时期肝活检组织中的survivin与p21waf-1基因和蛋白表达。结果:①HCC组织中survivin阳性率41.67%、癌旁肝组织54.17%;未发生肝癌及对照组大鼠活检肝组织,均未检出survivin蛋白(P<0.01);②HCC及癌旁肝组织中survivin mRNA的表达水平显著上调(P<0.01);③HCC及癌旁肝组织p21waf-1蛋白阳性率明显低于58周未出癌及对照组大鼠肝组织(P<0.01);④出癌大鼠p21waf-1 mRNA表达水平在HCC形成过程逐渐下调(P<0.01);⑤在HCC形成中,survivin与p21waf-1表达呈负相关。结论:在HCC形成的早期p21waf-1 mRNA表达的下调逐渐失去抑制细胞增殖、促进细胞分化功能,使细胞增殖,而survivin mRNA表达上调,抑制细胞凋亡,两者在HCC形成过程中可能发挥协同作用。 Objective:This investigation was to observe the expressions and the correlation of survivin and p21waf-1 genes in hepatocellular carcinogenesis. Methods: Survivin and p21waf-1 genes and proteins were detected by RT-PCR and immunohistochemistry in hepatocellular carcinomas (HCC) and liver tissues in rats. Results: OPositive rates of survivin protein in HCC and HCC-adjacent tissues were 41. 67% and 54. 17%, respectively. No survivin protein was found in biopsy and control animals liver tissues. (2)The expression of survivin mRNA was significantly up regulated in HCC and HCC-adjacent tissues. (3)Positive rates of p21waf-1 protein in HCC and HCC-adjacent tissues were significantly lower than those in biopsy and control animals liver tissues. (4)The levels of p21waf-1 mRNA in HCC developed rats were down regulated gradually and were significantly lower than those in biopsy and control animals liver tissues. (5)There was negative correlation between survivin and p21waf-1 in Hepatocellular carcinogenesis. Conclusion:The lower expression of p21waf-1 and over-expression of survivin may take a synergistic role in the course of hepatocarcinogenesis in rats.
