大鼠脑缺血再灌注后NF-κB的表达与神经细胞凋亡的关系 被引量:6

摘要 目的探讨大鼠脑缺血再灌注后脑组织中核因子-кB(NF-кB)表达水平与神经细胞凋亡的关系。方法采用大脑中动脉线栓法制备局灶性脑缺血再灌注模型,参考Longa的5分制法在大鼠麻醉清醒后进行评分,应用免疫组化、TUNEL法检测大鼠脑缺血再灌注后NF-кB的表达及神经细胞凋亡。结果大鼠脑缺血再灌注后缺血脑组织中NF-кB的表达明显增加(P<0.01),于24 h达到高峰,且神经细胞凋亡数明显增加(P<0.01)。结论脑缺血再灌注后神经细胞NF-кB表达水平增加,并参与神经细胞凋亡机制。 Objective To investigate the relationship between nuclear factor-кB (NF-кB) expression and neuron apoptosis after cerebral ischemia reperfusion injury in rats. Methods Rats were subjected to cerebral ischemia reperfusion injury through the occlusion of the middle cerebral artery. Neurological deficits were determined by Longa's score. Immunohistochemistry and TUNEL were used to detect NF-кB expression and neuron apoptosis. Results The expression of NF-кB in the ischemic area up-regulated after cerebral ischemia (P 〈0.01), peaking at 24 h after ischemia reperfusion injury. The number of apoptotic cells increased obviously (P 〈0.01). Conclusion The increase in the number of apoptotic cells is correlated with the up-regulation of NF-кB expression.
