乐昌含笑组织培养根的诱导及其POD和PPO活性的变化 被引量:3

摘要 以乐昌含笑组织培养中生长一致的未生根试管苗为材料,采用6-苄氨基嘌呤(6-BA)、萘乙酸(NAA)、吲哚丁酸(IBA)三因素二次正交旋转组合设计进行根的诱导培养,对乐昌含笑组织培养过程中根的诱导进行了3种生长调节剂不同配比的对比试验,观测乐昌含笑组织培养过程中根的诱导和形态的变化,测定每个试验组合的过氧化物酶(POD)活性和多酚氧化酶(PPO)活性的变化。结果表明:3种生长调节剂浓度过高或过低都不利于根的生长,当生长调节剂浓度为6-BA0.40mg·L-1,IBA0.60mg·L-1,NAA0.80mg·L-1时,生根状况最好。植物生根过程中,当植物体内POD活性变化先下降再升高后又下降而PPO活性变化先上升再下降后又上升时有利于生根。 Effects of 6-BA, NAA and IBA at various concentrations on the root induction of Michelia chapensis Dandy were investigated through three-element quadric orthogonal whirling combination design. Activities of peroxidase (POD) and polyphenol oxidase(PPO)were measured during the development of adventitious roots ,and also the root induction and patterns changes. The results showed that too low or too high of the concentration of three kinds of growth regulators was not conducive to the growth of root. And the suitable concentrations of growth regulators for rooting was 6-BA 0.40 mg·L^-1,IBA 0.60 mg·L^-1, NAA 0.80 mg·L^-1. POD activity decreased then increase then decreased and PPO activity increase then decreased then increase in Michelia chapensis Dandy were suitable for the root growth.
