摘要
以青钱柳离体胚为材料,采用正交试验设计方案对影响其萌发和成苗的4种因素进行了研究。结果表明:低温(4℃)层积有利于青钱柳离体胚的萌发,基本培养基种类是影响青钱柳离体胚萌发和生长的关键因素,低温层积后的离体胚适宜培养基组合为改良MS+6-BA 1.0 mg/L+NAA0.5 mg/L、萌发率达97.5%,适宜成苗的培养基组合为改良MS+NAA0.5mg/L、成苗率达93.33%,而6-BA对离体胚成苗则有抑制作用。
The relation was researched of germination rate and seedling percen of Cyclocarya paliurus Iljinsk in isolated seed embryo with 4 kind of factors by nrthogonal designing. The results showed that cold stratificatio was propitious to germinate of Cyclocarya paliurus Iljinsk in isolated seed embryo,the basal medium played a determinative role in the embryo germination and seedling development. The best media was modified MS+6-BA1 mg/L+NAA0.5 mg/L to promote germination of cold stratifieatio embryo, on which the pereentage of germination was up to 97.5%, and the optimal media to develop into seedlings was modified MS+NAA0.5 mg/L,the rate was 93.33%. 6-BA could inhibit the development of seedlings.
