摘要
[目的]研究人参皂甙Rb1对应激大鼠肝脏HSP-70(热休克蛋白质70)和PKC(蛋白激酶C)的影响及两者的关系。[方法]56只SD(Sprague-Dawley)大鼠分为人参皂甙组、生理盐水组、假手术组,均采用大鼠肝脏缺血再灌注制作应激模型,手术后于30min、60min及180min定点处死实验动物,收集肝脏标本,应用ELISA法测定大鼠肝细胞HSP70的表达及PKC的水平。[结果]人参皂甙30min组和60min组与生理盐水组比较,HSP70表达明显较高(P<0.05);人参皂甙60min组PKC水平较生理盐水组高,有显著性差异(P<0.05);大鼠肝脏的PKC水平与HSP70的表达进行线性回归分析显示两者呈显著的正相关。[结论]人参皂甙Rb1可以提高应激时大鼠肝脏HSP70的表达和PKC水平提高,且HSP70的表达与PKC水平的变化呈正相关。
[Objective] To study the effect of ginsenoside Rbl on liver heat shock protein 70 and protein kinase C and the relation-ship between the two factors. [Methods] 56 SD(Sprague Dawley) rats were divided into 3 groups: ginsenoside group, saline group, sham-operated group. Put these rats to death at 30min, 60rain and 180rain, extract liver and draw blood, use the ELISA to analyze the expression of HSP-70 and the level of PKC. [Results] (1) To compare the level of PKC at different time points of ginsenosides and the saline group, 60 rain ginsenoside group is higher than the saline group. There is a significant differenee(P〈0.05). Comparison of ginsenosides and saline 30 rain and 180 min is no significant difference(P〈0.05). (2)To compare HSP70 at different time points of ginsenosides and the saline group, 30 min and 60 min ginsenoside groups are higher than the saline group. There is a significant differenee(P〈0.05). Comparison of ginsenosides and saline 180 vain is no significant difference(P〉0.05). (3)The linear regression analysis of the level of PKC and the expression of HSP-70 show that two factors are in significant positive correlation. [Conclusions] 1. Ginsenoside Rbl can increase the expression of HSP70.2. Ginsenoside Rbl can increase the level of PKC. 3. The movement of the expression of HSP-70 and the level of PKC are in significant positive correlation.
