丙泊酚对大鼠肠缺血再灌注损伤时肝细胞凋亡及Caspase-3的影响

摘要 目的探讨丙泊酚对大鼠肠缺血再灌注损伤时肝细胞凋亡及Caspase-3的影响。方法成年健康Wistar大鼠42只,随机分为3组:实验对照组(C组,6只)、肠缺血再灌注组(IR组,18只)、丙泊酚干预组(P组,18只)。各组根据再灌注时间分为1,3,6h三个时相。通过夹闭肠系膜上动脉制作肠缺血再灌注损伤模型,实验结束后即刻取肝左叶做标本。用免疫组织化学法检测Caspase-3蛋白表达量,末端转移酶标记技术(TUNEL)法检测肝细胞凋亡指数(AI)。结果与C组比较,IR组和P组肝组织Caspase-3蛋白含量、AI均增加(P<0.05);与IR组比较,P组Caspase-3蛋白表达减少、AI减少(P<0.05)。结论丙泊酚可以下调大鼠肠缺血再灌注时Cas-pase-3蛋白表达,使肝细胞凋亡减轻,从而对肝损伤有一定的保护作用。 Objective To investigate the effects of propofol on hepatic cell apoptosis and expression of Caspase-3 with intestinal isehemia-reperfusion injury in rats. Methods Forty-two Wistar rats were randomly divided into 3 groups:control group(C group, n=6),isehemia-reperfusion group(IR group, n=18),propofol group(P group, n=lS).The rats were executed in each group after reperfusion for 1 h,3 h and 6 h respectively. A rat model of intestinal ischemia- reperfusion was established by blocking the superior mescnteric artery with clap. The left hepatic lobes were sampled at the end of reperfusion. Caspase-3 protein expression in liver tissues was assessed by immunohistochemical method. The apoptosis index(AI)of hepatic cell was measured by TUNEL method. Results Caspase-3 protein expression and AI of hepatic cell were significantly higher in IR group and P group than in C group(P〈0.05). Caspase-3 protein ex- pression and AI of hepatic cell were significantly lower in P group than in IR group (P〈0.05). Conclusions Propofot can relieve the liver cellular apoptosis by decreasing Caspase-3 protein expression. It may protect liver from intestinal isehemia-reperfusion injury.
