SPARC (secreted protein, acidic and rich in cysteine) is a key determinant of progressive fibrotic diseases. In vitro and in vivo studies have demonstrated that SPARC influences a variety of cellular activities. SPARC and its related peptides bind to several proteins of extracellular matrix (ECM), affect ECM protein expression, alter cell shape, reduce cellular adhesion, influence cell migration, and modulate growth factor-induced cell proliferation and angiogenesis. It is closely related to the occurrence and progress of fibrotic diseases. In view of the important roles of SPARC in a variety of fibrotic diseases, this paper presents the relationship of SPARC to systemic sclerosis, wound healing and pathological scar and its mechanisms.
