右归丸对脾肾两虚证大鼠甲状腺激素及环核苷酸水平的影响 被引量:10

Effects of Yougui Wan on Thyroid Hormone and Cyclic Nucleotides Levels in Rats with Spleen-kidney Deficiency Syndrome

摘要 【目的】通过观察甲状腺激素和环核苷酸水平的变化,探讨右归丸对实验性脾肾两虚证大鼠的防治作用,并从微观角度阐释脾肾之间的联系。【方法】选用SD雄性大鼠,随机分为正常对照组、模型组、右归丸组。前期采用大黄液灌胃(剂量为4 g/只)复制脾虚模型,后期采用氢化可的松肌肉注射(剂量为25 mg.kg-1.d-1)复制肾虚模型,分别给予蒸馏水、右归丸(剂量为5.8 g.kg-1.d-1)灌胃。观察动物的一般状态并测定血浆环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)及血清三碘甲状腺原氨酸(T3)、甲状腺素(T4)、反三碘甲状腺原氨酸(rT3)、促甲状腺素(TSH)的含量。【结果】模型组大鼠血清T3、T4、rT3含量在脾虚和脾肾两虚时均显著降低(P<0.01),血浆cAMP含量及cAMP/cGMP比值在脾虚和脾肾两虚阶段呈上升趋势(均P<0.01),cGMP含量在脾虚时无显著变化(P>0.05),至脾肾两虚时则出现降低(P<0.05)。而右归丸组可提高两个阶段T3、T4、rT3含量(P<0.05或P<0.01),前期脾虚时对环核苷酸无显著影响(P>0.05),后期可调节cAMP、cGMP含量及其比值(均P<0.01)。【结论】右归丸能够综合调整甲状腺轴激素含量和cAMP/cGMP比值从而防治脾肾两虚证。 Objective The changes of thyroid hormone and cyclic nucleotides levels in rats with spteen-kidneyy deficiency syndrome were observed to evaluate the preventive and therapeutic effect of Yougui Wan (YW) for spleen-kidney deficiency syndrome, and to explore the relationship between the spleen and the kidney at microscopic level. Methods Male SD rats were randomly divided into normal control group, model group, and YW group. Rat models of spleen-kidney deficiency syndrome were established by gavage firstly with rhubarb (4g for each rat) decoction and then with hydrocortisone (25 mg ·kg^-1·d^-1 , im). The model group and YW group were treated with distilled water and YW ( 5.8 g ·kg^-1·d^-1 ) respectively. The general status of rats was observed, and the plasma cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) levels as well as serum thyroid hormones (T3, T4 and rT3) and thyroid-stimulating hormone (TSH) contents were determined in all rats by radioimmunoassay (RIA). Results Compared with the normal control group, the serum contents of T3, T4, rT3 were decreased significantly ( P 〈 0. 01 ) , and plasma cAMP content and cAMP/cGMP ratio were increased significantly ( P 〈 0. 01 ) in rats with spleen deficiency and with spleen-kidney deficiency. cGMP content remained unchanged in rats with spleen kidney, but decreased in rats with spleen-kidney deficiency (P 〈 0.05). YW increased the content of T3, T4 and rT3 (P 〈 0. 05 or P 〈 0.01 ) in rats with spleen deficiency and with spleen-kidney deficiency. For rats with spleen deficiency, YW had no effect on their cyclic nucleotides levels, but decreased the contents of cAMP and cGMP and their ratio when the rats developed into spleen-kidney deficiency (P 〈 0.01 ). Conclusion The preventive and therapeutic mechanism of YW for spleen-kidney deficiency syndrome may be related with adjustment of thyroid hormones level and eAMP/cGMP ratio.
