摘要
目的探讨转化生长因子(TGF)-β1在系统性红斑狼疮(SLE)患者中的表达和临床意义。方法采用双抗体夹心酶联免疫吸附试验(ELISA)检测12例SLE血小板减少患者、9例血小板正常患者和12名健康对照者血清中的TGF-β1水平。SLE病例,用光学显微镜分类计数标准化骨髓涂片中的巨核细胞。结果SLE血小板减少组血清中TGF-β1浓度(3.63~32.84μg/L,中位数为10.64μg/L)和SLE血小板正常组患者血清中TGF-β1浓度(2.43~26.51μg/L,中位数为18.03μg/L)均减少,低于正常对照组(11.95~39.51μg/L,中位数为27.53μg/L),差异有统计学意义(P<0.05);SLE伴血小板减少组骨髓涂片均为巨核细胞计数正常或增多,颗粒巨核细胞增多,产血小板巨核细胞减少,伴有明显的巨核细胞成熟障碍。结论TGF-β1没有直接参与SLE血小板减少患者巨核细胞的负性调节,但TGF-β1作为一种强烈的免疫抑制因子,可能参与了SLE的发病机制。
Objective To discuss the clinical significance about detecting the plasma level of the factor TGF-β1 in the patients with systemic lupus erythematosus. Methods The plasma level of the factor TGF-β1 was detected in the 12 patients of thronthocytopenia, the 9 patients of platelet normal with SLE and 12 healthy volunteers by the ELISA kits. Results The plasma level of the factor TGF-I3t in the group of thrombocytopenia with SLE (3.63-32.84μg/L, median 10.64μg/L) and in the group of platelet nonnal with SLE (2.43-26.51μg/L, median 18.03 p.g/L), is lower than in the control group (11.95-39.51μg/L, median 27.53 p.g/L), and the result has statistical significance (P〈0.05); Bone marrow slides showed that megakary, ocytes counting generally are normal or increase, and granular pattern megalokaryocytes counting are increase, meantimely toc-platelet pattern megalokaryocytes counting are decrease, generally accompanying evident megakaryocytic dysmaturity in the patients of thrombocytopenia with SLE. Conclusion The plasma level of the factor TGF-β1 in SLE that belonging to active stage is lower.
