摘要
为确定柑桔皮中总黄酮的最优提取条件,提高提取得率,在单因素试验的基础上,采用三元二次回归正交旋转组合设计,研究在自然pH条件下微波功率、液料比、微波处理时间对黄酮产量的影响,建立各因子与黄酮产量关系的数学回归模型。试验结果表明:最佳提取条件:微波功率576W、液料比48∶1、微波处理时间194s。经试验验证,在此条件下提取2次,黄酮得率(3.38±0.09)%(n=3),与理论计算值3.490735%基本一致。回归模型能较好地预测柑桔皮中总黄酮的提取得率。
The aim of this work is the optimization of the extraction technics of total flavonoids from citrus peel by microwave-treatment. The effects of microwave power, ratio of solvent to material and treatment time were investigated at natural pH by using statistical techniques such as quadratic orthogonal rotary composite design which based on single factor experiments. The empirical model developed by response surface methodology (RSM) was adequate to describe the relationships between the studied factors and the response of flavonoid extraction yield. Based on the canonical analysis, the optimal conditions for maximizing flavonoid yield were treated for 194 s, at microwave power 576 W and ratio of solvent to material 48:1. Under the best extraction conditions whose frequency of extraction was 2, the yield of flavonoids was (3.38±0.09)%(n=3), which was close to the estimated value 3.490735% attained by using regression model. The model for the yield of total flavonoids had better prediction effects.
