摘要
单核细胞增生性李斯特氏菌是一种能够引起人畜共患疾病的食源性致病菌。为了鉴定该菌,以单核细胞增生性李斯特氏菌的李氏溶血素基因(Hly)为靶基因,通过环介导等温扩增技术(LAMP)对该基因进行扩增。共对12株菌进行了LAMP检测,结果表明:单核细胞增生性李斯特氏菌为阳性,其它菌株均为阴性。采用LAMP对单核细胞增生性李斯特氏菌纯培养进行检测,其方法的灵敏度为7.3×101cfu/mL;而利用LAMP检测人工污染的鸡肉中的单核细胞增生性李斯特氏菌,检出限为8.9×101cfu/g。
Listeria monocytogenes is a kind of food-borne pathogenic bacteria which can cause human and animal diseases. In order to detect Listeria monocylogenes, we ehosed the hemolysin gene (Hly)of L.monocytogenes as target gene, and amplified the gene by Loop-mediated isothermal amplification (LAMP). There were 12 bacterial strains to be detected by LAMP. The result of L. monocytogenes was positive and those of other strains were negative. With pure culture fluid as samples, the detection sensitivity of L.monocytogenes was 7.3×10^1 cfu/mL by LAMP. With artificially polluted chicken as samples, the detection limit of L.monocytogenes was 8.9×10^1 cfu/g by LAMP.
