PhaB和FQ-PCR两种快速检测结核分枝杆菌方法的比较

摘要 目的比较裂解型分枝杆菌噬菌体检测技术(Phage amplified biologically assay,PhaB)和荧光定量聚合酶链反应(FQ-PCR)快速检测临床标本中结核分枝杆菌的优缺点及临床应用价值。方法对142例肺结核患者的痰标本和41份非结核性痰标本应用PhaB法和FQ-PCR进行检测和结果比较。结果PhaB法阳性76例,阳性率53.5%;FQ-PCR法阳性84例,阳性率59.1%。PhaB法和FQ-PCR法相比,有统计学差异(P<0.05)。2种方法检测41份非结核性痰标本均为阴性。结论两种方法能快速、简便、灵敏、特异检测结核分枝杆菌,同时又有不同的特点。 Objective To compare the sensitivity and specificity by using mycobacteriophage splitting assay (PhaB) and fluorescence quantitative PCR (FQ-PCR), and evaluate the application value in clinical detection of Mycobacterium tuberculosis. Method The 142 sputum specimens of pulmonary tuberculosis, 41 sputum specimens of non-tuberculosis were detected by PhaB and FQ-PCR assay; the obtained results were compared each other. Rseults Of the 142 sputum specimens of pulmonary tuberculosis, 76 (53.5%) cases were confirmed as positive by using PhaB, 84 (59.1%) cases positive by FQ-PCR assay. The statistically significant difference of positive rate was found between PhaB and FQ-PCR assay (P〈 0.05); 4l sputum specimens of nontuberculosis were confirmed as negative by the two assays. Conclusion The PhaB and FQ-PCR assays have high sensibility and speeifietity, but each has it-self different dominance.
