酶解牡蛎肉制备高F值寡肽的研究 被引量:18

摘要 对采用胰酶水解近江牡蛎(Ostrea rivularis Gould)制备高F值寡肽进行了研究。其方法是将所得酶解液用活性碳吸附,然后用凝胶层析纯化。通过对比6种粉状活性碳去除酶解液中芳香族氨基酸和保留支链氨基酸的能力,找出了较适宜的活性碳类型及其吸附条件:活性碳型号为TX-325型,固液比为1:30(m/V),pH=2.5,吸附时间为3h,吸附温度为25℃。将经活性碳吸附后的酶解液用凝胶(型号为Biogel P2)层析法在流速为0.4mL/min的条件下进行分离纯化,可得到F值大于20的寡肽混合物,其相对分子质量约1450~800。 The meats of oysters were hydrolyzed by pancreatin and then the hydrolysate was treated by active carbon and gel filtration chromatography to prepare oligo-peptides with a high Fischer ratio. The aromatic amino acids adsorption and branched amino acids retention capacities of six kinds of active carbon were compared. Active carbon Biogel P2 was shown to be the best one and the best adsorption conditions with Biogel P2 were as follows: solid-liquid ratio 1:30, pH 2.50, adsorption time 3h, and obsorption temperature 25℃. The hydrolysate treated by Biogel P2 was further purified by gel filtration chromatography, giving the final product with high Fischer ratio of 〉20, and the molecular weights of800-1450 Da.
