摘要
采用Plackett-Burman试验设计法及响应面法分析,对产麦角固醇酵母菌株YF2-8进行了发酵条件优化研究。首先利用Plackett-Burman试验设计筛选出影响麦角固醇产量的三个主要因素:葡萄糖、蛋白胨、摇床转速。在此基础上用最陡爬坡路径逼近最大响应区域,再利用Box-Behnken试验设计及响应面分析法进行回归分析。结果表明,葡萄糖、摇床转速与麦角固醇产量有显著的相关性,并得到优化条件为:葡萄糖8.97%、蛋白胨2.36%、摇床转速为218r/min,麦角固醇产量为6.56×10-2mg/mL,麦角固醇产量提高了5.98%。
Fermentation parameters for ergosterol production by Saccharomyces cerevisiae YF2-8 were optimized by the method of Plaekett-Burman design and Response Surface Methodology. By the initial screening method of Plackett-Burman, the important factors affecting ergosterol production were shown as glucose, peptone and the rotating speed. Path of steepest ascent was undertaken to approach the optimal region of the significant factors. Box-Behnken design and Response Surface Methodology showed that glucose and shaking speed had an individually significant influence on ergosterol production. The ergosterol production was up to 6.56× 10^-2 mg/ml under optimal conditions (glucose 8.97%, peptone 2.36%, and the rotating speed 218 r/min), which was promoted by 5.98%.
