摘要 碱茅幼穗在附加有24-D15mg/L的MS培养基中,暗培养条件下两周时形成05cm2大小的愈伤组织块,其发生率为43%。培养3周后形成旺盛生长的肉黄色愈伤组织。愈伤组织的分化是在含KT10mg/L、NAA05mg/L的MS培养基上2~4周时出现绿色芽点,继续培养1~2周即可得到分化植株,将该植株移至不含激素的MS或N6培养基上时,光照条件下形成绿色分化植株。 The young spikelet of China Stargrass,Puccinellia distans in our experiment,when treated with aditional cultural medium of MS,had ever formed healed callus tissue by applying 2,4-D 15 mg/litre for 2 weeks under culturing conditions,and the rate of frequency was estimated at 43%. And if the culturing trial be lengthened longer than 3 weeks,then the healed tissue would be formed in vigorous vitality with flesh-yellowish color.Further on,whenever the differentiation of healed tissue was done on by applying KT 1.0 mg/litre and NAA 0.5 mg/litre on the basis of MS culture by lenghthening the trial time as long as 2 to 4 weeks,the tissue culture would appear the sprout point in green color;and if the culturing work would be gone on further for one to two weeks,then the differentiated plant tissues would be obtained at last.At this critical moment,when the plant was transformed on the culture medium MS without hormones or N6,under the condition of photosynthesis,the plant would be formed finally into greenized differentiated one.
