亚麻微生物脱胶技术的研究 Ⅲ.脱胶菌株适宜生长繁殖条件的测试 被引量:3

摘要 对两脱胶菌株Ke34及Le10的生长繁殖条件进行了测试。结果表明,它们的适宜扩繁培养基以豆饼粉培养基为最佳、其次为麦芙皮培养基,其中豆饼粉的含量以3%为宜;最适培养温度为35~40℃之间;最适pH值为70(中性);但是,以3%的豆饼粉培养基,pH值取其自然,培养效果也很理想。在优选各项测试的条件下,测定它们的生长曲线,并结合涂片镜检观察菌体变化,最终确定两脱胶菌株Ke34及Le10,以3%的豆饼粉为扩繁培养基、pH值70或取其自然,在有氧的条件,培养16~24h即为接种沤麻脱胶的最佳时间。 おwo degumming strains (Ke34 and Le10) had been tested for their growing and propagating in different media and under different conditions The results showed that the most suitable media for large scale-propagation was soy-bean cake powder media which was better than wheat bran media The fit content of soy-bean cake powder in media was 3% The most maderate temperature and pH for culture were 35~40℃ and 70, respectively Haver, the culture in the media with 3% soy-bean cake powder under non-adjnsted pH was also good According to their growth curve and observation through microscope, the media for a large scale propagation of the two degumming strains ke34 and Le10 contained 3% soy-bean cake powder The suitable pH volue for culturation was 70 or unadjusted Under the presence of oxygen, the best time to inoculate for degumming was between 16 and 24 hour after culturing
