摘要
研制了一款环介导等温扩增芯片,开发了相应的检测控制系统,并在此芯片上进行了金葡菌环介导等温扩增反应最佳扩增条件的研究.实现了在聚碳酸酯芯片上进行环介导等温扩增实验,与传统管式聚合酶链式反应相比,它需要的样品和试剂量更少.通过在不同温度下进行实验,获得了在芯片微体系条件下,进行环介导等温扩增反应检测金葡菌的最佳温度条件为61℃,提高了环介导等温扩增芯片的检测速度,缩短了核酸分析时间.
A section of loop-mediated isothermal amplification (LAMP) chip was introduced and its detecting platform was developed. Optimal DNA amplification of the detecting and controlling platform on the LAMP chip was studied. Using this chip and detecting platform, the lung infection bacterial staphyloccocus aureus rosebush was tested. Compared with conventional tubular polymerase chain reaction, more accurate results with smaller amount of samples and reagents were obtained by this chipbased LAMP analysis. The optimal experimental condition of isothermal DNA amplification of lab-on- a-chip (LOC) is obtained by the experiment, under which DNA detection of LOC is improved effectively and the time of DNA analysis is shortened.
