摘要
目的:探讨FHIT基因外显子5、8纯合性缺失与蛋白表达的关系及其在胃癌发病机制中的作用。方法:应用PCR及免疫组化SP法检测30例胃癌组织、18例正常胃组织中FHIT基因外显子5、8的纯合性缺失和FHIT蛋白的表达。结果:FHIT蛋白阴性表达率及外显子5、8纯合性缺失率在胃癌组织中均高于正常胃组织(P<0.05)。FHIT蛋白表达与胃癌的分化程度有关(P<0.05);胃癌组织中FHIT基因外显子5、8纯合性缺失与FHIT蛋白阴性表达有显著相关性(r=0.59,P<0.05)。结论:胃癌组织中FHIT基因外显子5、8纯合性缺失可能是引起FHIT蛋白表达降低并进而导致胃癌发生的重要机制。
Objective To investigate the homozygous deletion of FHIT gene exon5, 8 and the expression of FHIT protein in gastric carcinoma. Methods The homozygous deletion of FHIT gene exon5, 8 and the expression of FHIT protein were detected by PCR and immunohistochemistry S-P technique in 30 cases of gastric carcinoma specimens and 18 cases of normal gastric tissues. Results The negative expression rate of FHIT protein and the total homozygous deletion rates of FHTI gene exon5, 8 were significantly higher in carcinoma tissues than those in normal tissues (P 〈 0.05). Expression of FHIT protein was significantly correlated with tumor differentiation (P 〈 0.05). The correlation between homozygous deletion of FHTI gene exon5, 8 and the negative expression of FHTI protein in gastric carcinoma was statistically significant (r = 0.59,P 〈 0.05). Conclusion The homozygous deletion of FHIT gene exon5, 8 may be a major way of inducing the inactivation of FHIT gene and the carcinogenesis of gastric carcinoma.
