摘要
从银杏中分离出一株能产生银杏内酯类物质的内生真菌 Asprgillus fumigatusvar.fumigatus FG052,以其为出发菌株,经紫外和亚硝酸复合诱变,结合自身代谢产物的抗性筛选,获得一株遗传稳定性较好的变株 m_4-4,效价较出发茵株提高56%.对 FG052发酵条件优化表明,最适发酵培养基为:葡萄糖(3%),NaNO_3(0.3%),发酵起始 pH 值7.最适发酵条件为:接种龄为18h,接种量为1%,250mL 摇瓶的装液量为30mL,发酵温度为26℃.
An endophytic fungi Aspergillus fumigatus var. fumigatus FG052 strain producing ginkgolides was isolated from Ginkgo biloba L. ,and as an original strain, the endophytic fungi was mutated and bred by UV an nitrous acid combined treatment. A high yield mutant m4-4 was obtained by resistance screening with self metabolites,and its productivity was increased by 56% than the start strain.The results of experimenting on optimal fermentation conditions of FG052 strain showed that optimum culture medium for fermentation were shown as follows:NaNO3 0.3% ,glucose 3% ,pH 7.0 before sterilized. The optimum fermentation conditions were predicted as below: seed age 18 h, inoculated volume 1%, liquid culture volume 30 mL in 250 mL shake flask, fermentation temperature 26 ℃.
