摘要
目的:探讨大鼠胆道梗阻再通术后肾脏水通道蛋白3(AQP3)mRNA的变化。方法:50只大鼠,随机选10只作为对照组,其余为实验组。实验组先建梗阻性黄疸(obstructive jaundice,OJ),7d后解除梗阻。按取材时间不同分为5组,OJ0h、OJ24h、OJ72h、OJ1周和NC组。测定血生化指标,RT-PCR检测肾脏AQP3 mRNA。结果:随梗阻解除时间延长,总胆红素(TBIL)和丙氨酸氨基转移酶(ALT)逐渐下降(P<0.05);OJ0h与NC相比血尿素氮(BUN)、肌酐(Cr)无明显变化(P>0.05),AQP3 mRNA表达降低;术后24h(OJ24h)BUN、Cr下降,AQP3 mRNA表达进一步降低。OJ72h与OJ24h相比BUN、Cr无明显变化(P>0.05),AQP3 mRNA表达增强,但仍弱于NC组(P<0.05);到第7天,BUN、Cr和AQP3 mRNA均与NC无明显差异。结论:大鼠胆道梗阻解除后AQP3 mRNA的变化与AQP3的变化一致,其灵敏性和特异度均高于BUN、Cr。
Objective To explore the changes of renal AQP3 mRNA in rats after recanalization of bile duct occlusion. Methods The kidney samples used in this study were from the previous experiment, so were the biochemical parameters. The expression of AQP3 mRNA was detected by RT-PCR. Results Levels of TBIL and ALT were decreased gradually with the increasing duration of recanalization of the occluded bile duct (P 〈 0.05). As compared with the control subjects, levels of BUN and Cr did not differ significantly in the rats at the moment their bile duct was recanalized (P 〉 0.05), AQP3 mRNA expression was downregulated (P 〈 0.05) ; levels of BUN and Cr were increased on day 1 of recanalization (P 〈 0.05) and AQP3 mRNA expression was further declined (P 〈 0.05) ; on day 3, levels of BUN and Cr did not differ significantly from those on day 1 (P 〉 0.05), and AQP3 mRNA expression was upregulated but the expression level was lower (P 〈 0.05) ; on day 7, there were no significant differences in levels of BUN and Cr and AQP3 mRNA expression. Conclusions Following recanalization of bile duct occlusion, the change in AQP3 mRNA expression is similar to that in AQP3.
