ICG联合光照对兔晶状体上皮细胞增殖的影响 被引量:5

摘要 目的观察兔晶状体囊外摘除术(ECCE)中使用吲哚青绿(ICG)水分离联合光照对残留晶状体上皮细胞(LECs)增殖的抑制作用。方法40只兔随机分成ICG+光照组、ICG组、光照组和空白对照组,每组各10只兔(20眼)。各组均行ECCE术。ICG+光照组用0.5%ICG做水分离联合卤素灯光源照射;ICG组用0.5%ICG水分离;光照组用BSS水分离联合卤素灯光源照射;空白对照组用BSS做水分离。术后第1、7天时每组随机处死5只兔(10眼)。免疫组织化学方法检测各组LECs的增殖细胞核抗原(PCNA)表达,计算增殖指数(PI)。结果术后第1天时,ICG+光照组LECs呈单层排列,其他各组LECs呈单层或复层排列;术后第7天时,ICG+光照组LECs少或无,其他3组LECs呈多层束状或成簇排列。ICG+光照组术后第1天时,PI值(7.5±3.7)%,低于其他3组,差异均有显著性(P<0.05);术后第7天时ICG+光照组,LECs极少,ICG组、光照组及空白对照组PI值组间差异无显著性(P>0.05)。结论兔ECCE术中采用0.5%ICG水分离联合光照,可早期抑制LECs的增殖。 [Objective] To investigate the potential of ICG combined with visible light in inhibiting LECs in vivo that are left in the capsular bag after extracapsular cataract extraction (ECCE) in a laboratory setting using albino rabbits. [Methods] 40 albino rabbits were randomly assigned to 4 groups: ICG+light group, ICG group, light group and control group. Eaeh group had 20 eyes from 10 rabbits. ECCE was performed in each group. When performing hydrodissection, ICG+light group: hydrodisseetion using 0.5% ICG eombined with light; ICG group: hydrodisseetion using 0.5% ICG ; light group: hydrodisseetion using BSS combined with light; control group: hydrodissection using BSS. 5 rabbits were saerifieed using randomization at postoperative day 1 and 7 respectively. The expression of PCNA in LECs was detected by immunohistoehemistry, and proliferation index (PI) was calculated for each group. [Result] At pest-operative day 1, LECs were aligned in single layer in ICG+light group, while in single or multi-layer in the other three groups. At pest-operative day 7, none or only few LECs were found in ICG+light group, while LECs became multi-layer with fusiform and sliver eells and polygon eells in the other 3 groups. PCNA expression of LECs in each group at different time points: at post-operative day 1, PI was 7.5(3.7% in ICG+light group, significantly lower than the other three groups (P 〈0.05), while there were no significant differenees in PI among the other 3 groups (P 〉0.05). At post-operative day 7, PI was not ealeulated in ICG+light group due to insuftieient cells, where as there were no signifieant differences in PI among the other 3 groups (P〉0.05). [Conclusion] ICG (0.5%) hydrodisseetion eombined with visible light inhibited LECs proliferation in early postoperative stage in experimental ECCE using albino rabbits.
