摘要
为研究新城疫病毒HN融合蛋白抗肿瘤细胞的作用机制,以马立克病肿瘤细胞系MDCC-MSB1为体外研究对象,将HN融合蛋白与MDCC-MSB1细胞共培养后,应用CCK-8法检测HN融合蛋白对肿瘤细胞的增殖抑制作用,用琼脂糖凝胶电泳分析DNA断裂程度,用流式细胞仪检测肿瘤细胞的凋亡情况。结果表明,HN融合蛋白对MDCC-MSB1肿瘤细胞的增殖有抑制作用,并呈时间、剂量依赖性;DNA出现断裂现象;MDCC-MSB1细胞出现凋亡。证实,HN融合蛋白体外可诱导MDCC-MSB1肿瘤细胞凋亡。
MDCC- MSBI(MD lymphoblastoid cell line) was used in vitro to investigate the anti tumor effect of HN fusion protein of Newcastle disease virus. After MDCC MSB1 cells were treated with HN fusion protein for some time,the inhibitive effect of HN fusion protein on MSB1 cell was detected by CCK-8 assay,agarose gel electrophoresis and flow cytometry analysis. HN fusion protein inhibited the growth of MDCC-MSB1 cells,caused the fractures of DNA in MDCC MSB1 cells and the apoptosis of MDCC-MSB1 cells,and this effect was dose- and time-dependent. The results showed that HN fusion protein induced apoptosis of some MDCC-MSB1 in vitro.
