摘要 目的探讨一种低浓度红细胞同种抗体浓缩提纯的方法。方法采用液态氯化钙-草酸钾法("钙-草"法)或凝血酶法去除血浆中纤维蛋白,使血浆转化成血清,再经聚乙二醇(PEG)法浓缩提纯。对浓缩提纯的终产品进行各项质量指标检测及初步应用。结果处理的8份抗-M、N、S血浆("钙-草"法)、4份抗-D、E血浆("钙-草"法)及15份抗-A、B血浆(凝血酶法和"钙-草"法)成功去除了血浆中的纤维蛋白,转化成血清,浓缩提纯的终产品各项质量指标均合格,检测献血者红细胞和血型的结果与市售试剂一致。结论以"钙-草"法去除血浆纤维蛋白,PEG法浓缩提纯血型抗体操作简便,成本低,便于推广使用。 Objective To develop a method for concentration and purification of low concentration red cell alloantibody. Methods The fibrin in plasma was removed by liquid calcium chloride-potassium oxalate method or by thrombin, and the prepared serum was concentrated and purified by PEG. The final product was subjected to control tests and applied primarily. Results All the 8 anti-M, anti-N and anti-S plasma samples, 4 anti-D and anti-E plasma samples as well as 15 anti-A and anti-B plasma samples were successfully prepared into sera by removal of fibrin. The final products were qualified in control tests, by which the test results of red cells and blood groups of donors were consistent with those by commercial reagents. Conclusion The preparation of blood group antibody by removal of fibrin from plasma by liquid calcium chloride-potassium oxalate method as well as concentration and purification with PEG was simple and easy to be popularized.
