摘要
目的探讨甲状旁腺激素相关蛋白基因(parathyroid hormone-like hormone,PTHLH)mRNA在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)黏膜及正常口腔黏膜中的表达及意义。方法选取30例OSCC患者,逆转录聚合酶链反应(reverse transcription-PCR,RT-PCR)方法检测肿瘤组织和自身对照正常口腔黏膜中PTHLH mRNA的表达,SAS6.12统计软件进行χ2检验和t检验。结果PTHLH mRNA在OSCC黏膜和自身对照正常黏膜中表达的阳性率分别为83.0%和36.7%,差异有统计学意义(χ2=11.74,P<0.001)。PTHLH mRNA在OSCC黏膜和自身对照正常黏膜中的表达水平分别为2.38±0.15和0.74±0.10,上调3.02倍,差异有统计学意义(t=49.83,P<0.01)。PTHLH mRNA在OSCC中表达上调与肿瘤临床分期(t=1.60)、病理分化程度(t=1.95)、是否发生颈淋巴结转移(t=1.73)无关,差异均无统计学意义(P>0.05)。结论OSCC癌变过程中PTHLH mRNA表达水平显著上调,PTHLH基因可能是OSCC的1个靶基因,在OSCC分类诊断中有应用价值。
Objective To investigate the mRNA expression of PTHLH gene in oral squamous cell carcinoma (OSCC) and paired normal tissue. Methods Differential PTHLH gene expression between 30 eases of paired normal and eancerous tissues were deteeted by reverse transcription-PCR (RT-PCR). The data were analyzed with SAS 6.12 software for Chi-square and t test. Results RT-PCR results demonstrated that the positive rate of PTHLH mRNA in 30 eaneerous tissues and paired normal samples were 83.0% (25/30) and 36.7% ( 11/30 ) respectively ( P 〈 0.001 ) ; relative expression level of PTHLH mRNA in 30 eancerous tissues and paired normal samples were 2.38 ± 0.15 and 0.74 ± 0.10 respectively ( P 〈 0.01 ), with a up-regulating of 3.02-folds. Additionally, no significant difference of PTHLH mRNA level was found among different clinical stages, grades of tumor differentiation and with or without lymph nodes metastasis ( P 〉 0.05 ). Conclusion PTHLH gene might play a important role in OSCC carcinogenesis and could be a potential target of OSCC therapy.
