雷公藤甲素对大鼠脑皮质Fas蛋白表达的影响及时间节律性研究 被引量:6

摘要 目的:本实验观察雷公藤甲素对大鼠脑皮质Fas蛋白表达的影响并探讨其时间节律性,为临床合理用药提供实验数据和理论依据。方法:雄性SD大鼠40只随机分为4组,包括空白对照组;雷公藤甲素Ⅰ组20μg/kg,bid(8am,8pm);雷公藤甲素Ⅱ组40μg/kg,qd(8am);雷公藤甲素Ⅲ组40μg/kg,qd(8pm)。连续灌胃7周后断头处死,用免疫细胞化学染色法观察大鼠脑皮质Fas蛋白的表达变化。结果:雷公藤甲素Ⅰ组大鼠脑皮质FAS蛋白的免疫反应的表达最高,雷公藤甲素Ⅲ组FAS蛋白的免疫反应仅次于雷公藤甲素Ⅰ组,而雷公藤甲素Ⅱ组仅可见少量表达;空白对照组未见大鼠脑皮质FAS蛋白的免疫反应。结论:雷公藤甲素可通过死亡受体介导细胞凋亡的途径诱导细胞凋亡,从而发挥其抗肿瘤、免疫抑制作用,且雷公藤甲素的作用具有时间节律性。 Objective:To observe the effect of Triptolide on the Fas protein expression and the temporal rhythm of the drug in the brain cortex of experimental rats for providing the data with safe use in clinic. Methods : Forty SD male rats were randomized into groups of the control ,Triptolide- Ⅰ20 μg/kg,bid (8 am,8 pm),Triptolide-Ⅱ 40 μg,/kg,qd(8 am) and Triptolide-Ⅲ 40 μg/kg,qd(8 pm). After 7 consecutive weeks of administration by gavage,the rats were decapitated for taking the cortex tissues by using immune cell staining technique to determine the changes of the Fas protein expression in the brain cortex. Results : The Fas protein ex- pression of immune response was the highest in group Triptolide-Ⅰ and Triptolide-Ⅲ ranked the second. Only a little expression was found in the group Triptolide-Ⅱ while the response was absent in the controls. Conclusion:Triptolide may induce the apoptosis through the death receptor-mediated factors to function in anti-tumor and immunosuppression. In addition, the drug action may be involved in the temporal rhythm.
