针刺预处理对大鼠脑缺血耐受及促红细胞生成素的诱导作用 被引量:10

摘要 目的探讨大鼠脑缺血及针刺预处理后促红细胞生成素(EPO)表达的变化及其意义。方法建立大鼠局灶性脑缺血及针刺预处理模型。随机分为对照组(缺血组)、针刺预处理+局灶性脑缺血组(n=8)。应用普通病理、免疫组化和原位杂交技术,观察大鼠脑缺血及缺氧预处理后EPO表达变化。结果脑组织中EPO蛋白及mRNA表达可见于存活和坏死的神经细胞、血管内皮细胞、胶质细胞。脑缺血诱导了EPO的表达,针刺预处理后EPO在缺血周边区表达更为明显(P<0.05)。结论针刺预处理后EPO在周边区表达增强,其表达可见于存活的神经细胞、血管内皮细胞及胶质细胞,提示EPO参与了针刺预处理的脑保护机制,其保护作用可能通过神经营养和血管生成双重作用完成。 Objective To explore the changes of erythropoietin (EPO) expression between cerebral ischemia and acupuncture preconditioning and its significance. Methods The rat models of focal cerebral ischemia and acupuncture preconditioning plus focal cerebral ischemia were established, 8 in each group. EPO expression in two groups was observed by general pathology, immunohistoehemistry and in situ hybrization. Results The expressions of EPO protein and mRNA in cerebral tissue were seen in the survival and apoptotic nerve cells, vascular endothelia cells and gliocytes. Cerebral ischemia induced EPO expression, especially in ischemia border area after acupuncture preconditioning( P 〈 0.05 ). Conclusions The enhanced EPO expression in ischemia border area 'after treated by acupuncture is in survival neural cells,vascular endothelial cells and gliocytes, suggesting that EPO participates in the cerebral protection mechanism of acupuncture preconditioning by neurothophy and angiogenesis.
