摘要
目的:探讨Hcy是否影响乳腺癌患者外周血MNC表达uPA、uPAR和PAI-1.方法:采用不同浓度Hcy对乳腺癌患者外周血MNC进行短期(4h)培养后,粉碎细胞,ELISA法检测其uPA,uPAR和PAI-1的表达.结果:Hcy诱导乳腺癌患者外周血MNC表达uPA,uPAR,而抑制MNC表达PAI-1,并且呈剂量依赖性,Hcy浓度为0,12.5,25,50,100μmol/L时,uPA分别为47.00±21.26,57.70±26.72,72.38±38.64,83.33±42.74,94.83±44.47 ng/L;uPAR分别为1299.3±291.7,1447.2±408.8,1607.6±467.0,1731.3±521.1,1815.3±522.3 ng/L;PAI-1分别为52.5±20.7,41.6±21.9,35.4±17.9,32.6±18.3,28.7±16.2μg/L.结论:Hcy可以影响乳腺癌患者外周血MNC表达uPA,uPAR和PAI-1,可能在乳腺癌的侵袭和转移中发挥一定的作用.
AIM: To explore whether homocysteine affects the peripheral mononuclear cells (MNC) in expressing urokinase-type plasminogen activator (uPA) , urokinase-type plasminogen active receptor (uPAR) and plasminogen activator inhibitor-1 (PAI-1) in patients with breast carcinoma. METHODS: The peripheral MNCs were incubated with euhure liquids of different Hcy concen- trations for 4 h and then were smashed, and the levels of uPA, uPAR and PAI-1 were measured by ELISA. RESULTS: Hcy induced the expression of uPA and uPAR but inhibited the expression PAI-1 in a dose-dependent manner. CONCLUSION: Hcy affects the peripheral mononuelear cells in expressing uPA, uPAR and PAI-1, and may play a role in the invasion and metastasis of the breast carcinoma.
