热休克反应对内毒素诱导的IP-10基因表达的影响

摘要 目的探讨在RAW264.7巨噬细胞中热休克反应(HSR)对大肠杆菌内毒素(脂多糖,LPS)诱导的干扰素诱导蛋白10(IP-10)表达的影响。方法RAW264.7巨噬细胞分别给予不同剂量及不同反应时间的LPS处理,并行HSR后抽提总RNA进行RT-PCR实验。结果LPS可促进RAW264.7巨噬细胞中IP-10基因的转录,具有剂量依赖性;用浓度为600μg/L的LPS刺激2h并行HSR时,IP-10的mRNA表达量最大。结论HSR能促进LPS诱导的IP-10基因mRNA表达水平。 Objective To explore the effects of heat shock response (HSR) on RAW264.7 macrophages interferon induced protein 10 (IP - 10) expression induced by lipopolysaecharide (LPS). Methods RAW 264.7 Macrophage cells were exposed to LPS with different doses and for different reaction time, and part of cells were treated with heat shock. Then total RNA were extracted respectively. The expression of IP- 10 were detected by RT-PCR. Results LPS could enhance the transcrition of IP - 10 in RAW264.7 macrophages, which showed dose reliability. When RAW264.7 macrophages were treated with LPS at concentration of 600μg/L and heat shock for 2h, IP- 10 rnRNA reached to a peak level. Conclusion HSR can promote the level of IP- 10 mRNA induced by LPS.
