AFFINITY PURIFICATION OF NERVE GROWTH FACTOR ANTIBODIES

通过神经生长因子(NGF)-Sepharose 4B 亲和层析柱从兔抗鼠 NGF 血清获得纯化的抗体。SDS 凝胶电泳表明其主要成分为 IgG。在体外它可以完全阻断 NGF 刺激鸡胚背根节长出突起的效应;注入新生鼠它可致免疫去交感作用。与抗血清比较,抗体约被纯化了10～15倍。

Pure antibodies to nerve growth factor (NGF) have been isolated from rabbit NGF an- tiserum by affinity chromatography.The affinity column (1.2×8.0cm) was prepared using mouse 2.5S NGF linked to Sepharose 4B and the coupled antibodies were eluted with high salt concentration (4.5M MgCl_2). It was found that the product is mainly IgG on SDS gel electrophoresis.The biological and immunological activities were tested by (1) blocking NGF activity in a tissue culture assay,and (2) immunosympathectomic effect.The result showed that NGF antibodies were purified approximately 10-15 folds. NGF-like immunoreactivity was observed satisfactorily in male mouse submandibular glands by application of purified antibodies.
