摘要
人外周血 T 克隆细胞在 YIL-2持续刺激下具有良好的体外扩增能力和LAK 活性。在半固体琼脂培养体系中,T 克隆细胞抑制骨髓 CFU-GM 形成,抑制效应与 T 细胞数量、T 细胞和骨髓细胞的预孵育时间呈正相关。自体及异体 T 克隆细胞对 CFU-GM 的抑制作用无显著差异。经 FACS 表型分析,这种 IL-2激活的 T 克隆细胞为T_3(+)、T_4(-)、T_8(+)、DR(+)、Tac(+)。
T cell clones from human PBL of healthy adults treated by recombinant interleukin-2 (rIL-2) showed inhibitory effect on myeloid progenitor cells growth when the cloned T cells were added into a semisolid medium with a shorter pre-incubation (0.5-4 hours) with bone marrow mononuclear cells (BMMNC).The degree of colony inhibition for GM was well related to the time of T-BMMNC interaction as well as to the number of cloned T cells added into cultures.No substantial difference of the inhibitory effect was found between autologous and allogenic T cell clones under same condition.These cloned T cells,with con- tinued rIL-2-stimulation,also showed either good ability of expansion in vitro or lymphokine- activated killer (LAK) activity against NK-sensitive erythroleukemic cell line K562 and NK- resistant human liver cancer cell line H7402 in a 4 hours ^(51)Cr release assay.Phenotypic analysis by flow cytometry indicated that these T clone cells carried T_3,T_8.Tac and DR antigens.These data suggest that cell-to-cell interaction can be the main way at least in the inhibitory effect of rIL-2-activated T clone cells on growth of myeloid progenitor cells,that may be involving cytotoxicity as well as LAK activity and contact inhibition.
