过表达小窝蛋白-1对293T细胞生长的影响及机制

摘要 目的:探讨小窝蛋白-1(caveolin-1)蛋白过表达对细胞生长与增殖的影响及其机制。方法:将重组的真核表达质粒PHLCX Nflag3/小窝蛋白-1以脂质体法转染293T细胞,流式细胞仪测定细胞周期变化;Western Blotting分析P53、P21、ERK、pp-ERK蛋白表达。结果:293T细胞出现了G0/G1期阻滞;P53/P21表达增加,ERK/ppERK表达减少。结论:过表达小窝蛋白-1抑制293T细胞生长,并且这种对细胞生长抑制作用通过P53/P21与ERK信号传导通路实现的。 Objective To investigate the effects of caveolin-1 overexpression on the growth and proliferation of 293T cells and its mechanism. Methods Recombinant plasmid PHLCX Nflag3/caveolin-1 was transfectod into 293T cell by liposome method,the cell cycle was detected by FCM; the expression of P53, P21, ERK and pp-ERK was detected with Western Blotting analysis. Results The overexpression of eaveolin-1 arrested 293T cells in G0/G1 phase of the cell cycle;the expression of P53, P21 increased and the expression of ERK and pp-ERK decreased. Conclusion Overexpression of caveolin-1 may inhibit the growth of 293T cells,P53/P21 and ERK signal transduction pathway may involve in the inhibitory effect.
