摘要
目的:研制基于蛋白A法的禽流感压电免疫芯片。方法:以AT切型、基频10MHz的石英晶体为材料,设计压电免疫芯片。采用蛋白A法将禽流感H5亚型单克隆抗体和抗原的反应液固定在石英晶体的金膜电极表面,构建可用于禽流感检测的压电免疫芯片。结果:蛋白A法若先固定抗体,将无法检测到抗原;但可先进行抗原抗体反应再与蛋白A结合以达到定性检测的效果。结论:蛋白A法可作为禽流感压电免疫芯片抗体固定的方法,但在应用方面存在一定的局限性。
Objective: To establish a piezoelectric immunosensor for avian influenza viruses by SPA method Methods:The mixture of H5 subtype AIV monoclonal antibody and antigen is deposited on the gold electrode's surface of 10 MHz quartz AT-cut crystal by SPA method to made the chips. Results: If the monoclonal antibody is immobilized to SPA, the antigen could not be detected; but the mixture of H5 subtype AIV monoclonal antibody and antigen can be detected and could be used to qualitative detection. Conclusions: The SPA method could be used to establish a piezoelectric immunosensor for avian influenza viruses, but there is limitation in use.
