糖尿病大鼠甲状腺组织TG,TPO,TSHR与RAGE mRNA表达的实验研究 被引量:2

Experimental investigation on TG, TPO, TSHR and RAGE mRNA expression in thyroid tissue of diabetic rats

摘要 目的探讨糖尿病大鼠甲状腺组织甲状腺球蛋白(TG)、甲状腺过氧化物酶(TPO)、促甲状腺激素受体(TSHR)与晚期糖基化终末产物受体(RAGE)mRNA表达的变化。方法Wistar大鼠112只,随机分为糖尿病组(DM组)27只、糖尿病氨基胍治疗组(AG组)28只、糖尿病胰岛素治疗组(INS组)32只和对照组(N组)25只,采用逆转录-聚合酶链反应检测造模后12和20周各组大鼠甲状腺组织TG、TPO、TSHR和RAGEmRNA的表达。结果TGmRNA表达:12周各组之间无统计学差异(P〉0.05)。20周DM组低于AG组(P〈0.01)、INS组(P〈0.01)和N组(P〈0.01)。后3组间无统计学差异(P〉0.05)。TPOmRNA表达:12周DM组低于AG组(P〈0.05)、INS组(P〈0.001)和N组(P〈0.001),AG组低于INS组(P〈0.001)和N组(P〈0.001),INS组低于N组(P〈0.01)。20周DM组低于INS组(P〈0.01)和N组(P〈0.001),与AG组无统计学差异,AG组低于INS组(P〈0.05)和N组(P:0.001),INS组与N组无统计学差异。TSHRmRNA表达:12周DM组高于AG组(P〈0.05)、INS组(P〈0.05)和N组(P〈0.01),后3组间无统计学差异(P〉0.05)。20周各组之间差别无统计学意义(P〉0.05)。RAGEmRNA表达:12周和20周均表现为DM组高于AG组(P〈0.001)、INS组(P〈0.01)和N组(P〈0.001),INS组高于AG组(P〈0.05)和N组(P〈0.01),AG组与N组无统计学差异。结论糖尿病大鼠甲状腺组织TGmRNA、TPOmRNA表达降低,TSHRmRNA表达增高,可能与高糖毒性所致甲状腺组织RAGEmRNA表达增高有关。 Objective To investigate the expression alteration of thyroglobulin ( TG), thyroperoxidase ( TPO), thyrotropin receptor(TSHR) and receptor for advanced glycated end products (RAGE) mRNA in the thyroid tissue of diabetic rats. Methods 12 weeks and 20 weeks after the animal model establishment, the TG,TPO,TSHR and RAGE mRNA expresssion in the thyroid tissue of diabetic rats( DM group), diabetic rats treated with aminoguanidine(AG group), diabetic rats treated with insulin(INS Group) and normal rats( N group) were detected with RT-PCR. Results TG mRNA expression:There were no statistical difference among each group of 12^th week(P 〉 0.05 ). But for 20^th week, DM group was lower than AG group (P 〈0.01 ), INS group ( P 〈 0. 01 ) and N group ( P 〈 0.01 ), but there were no statistical difference among the latter three groups (P 〉 0.05 ). TPO mRNA expression:For 12^th week, DM group was lower than AG group( P 〈 0.05 ), INS group (P〈0.001) and N group(P 〈0.001),AG group was lower than INS group(P 〈0.001) and N group(P〈 0.001 ) , INS group was lower than N group ( P 〈 0.0 1 ) . For 2 0th week , DM group was lower than INS group (P 〈0.01 ) and N group (P 〈 0. 001 ), but with no difference from AG group. AG group was lower than INS group (P 〈 0.05) and N group ( P = 0. 001 ), but there was no difference between INS group and N group. TSHR mRNA expression:For 12th week, DM group was higher than AG group(P 〈 0.05 ), INS group (P 〈 0.05 ) and N group ( P 〈 0.01 ), but there were no statistical difference among the latter three groups (P 〉 0.05 ). For 20th week, there were no statistical significance in the difference among each group(P 〉0.05). RAGE mRNA expression: for both of 12th and 20th week, DM group were higher than AG group ( P 〈 0. 001 ), INS group ( P 〈 0.01 ), and N group (P 〈 0.001 ), INS group were higher than AG group ( P 〈 0.05 ) and N group(P 〈0.01 ) ,there was no difference between AG group and N group. Conclusion In the thyroid tissue of diabetic rats,the decreased expression of TG mRNA,TPO mRNA and increased expression of TSHR mRNA may be related with the overexpression of RAGE mRNA that caused by hyperglycemia.
