摘要
目的:建立HPLC法测定蒲公英提取物中绿原酸含量。方法:采用Agilent Zorbax SB C18柱(4.6mm×250mm,5μm),流动相为乙腈-0.4%磷酸溶液(10∶90),流速1.0mL·min-1,检测波长324nm,柱温30℃。结果:绿原酸的线性范围为0.04~0.60μg(r=0.9992),平均加样回收率(n=5)为96.48%。结论:采用HPLC法测定蒲公英提取物中绿原酸含量的方法快速、简单、准确,可用于蒲公英提取物中绿原酸含量的测定。
Objective :To establish an HPLC method for the determination of chlorogenic acid in dandelion extract. Methods: Agilent Zorbax SB C18 column (4.6 mm× 250 mm, 5 μm) was adopted ; The mobile phase consisted of acetonitrile- 0. 4% phosphoric acid solution( 10: 90)at a flow rate of 1.0 mL ·min^-1; The detection wavelength was set at 324 nm,and column temperature was 30 ℃. Results:The linear ranges of chlorogenic acid was 0. 040 - 0. 60 μg( r = 0. 9992 ) ; The average recoveriey ( n = 5 ) was 96.48%. Conclusion: HPLC determination of chlorogenic acid in dandelion extract is rapid, simple and accurate. It can be applied to determine of chlorogenic acid in dandelion extract.
