摘要
以魔芋粉为诱导底物,分别从底物浓度、氮源的选择、氮源重组比例、氮源的浓度及接种量等方面进行乳白耙菌β-葡甘聚糖酶产酶条件优化,最后进行了产酶动力学分析。实验结果表明,最优的培养条件为:魔芋粉浓度为2%,以蛋白胨和硫酸铵为氮源,二者添加比例为1:2,混合氮源浓度为0.4%,接种量为10%。发酵4d,β-葡甘聚糖酶酶活达到48.1IU/mL,是未优化培养基条件下酶活的2.3倍。
The producing conditions of p-mannan mannohydrolase of Irpex lacteus were optimized, including concentration of konjac power, choice of nitrogen resource, recombinant proportion of nitrogen resource, concentration of nitrogen resource and innoculation volume,at last the producing-enzyme kinetic was analyzed. The optimization conditions were obtained as follows: konjac power concentration was 2%, and peptone and ammonium sulfate were choose as nitrogen resource, and the recombinant proportion was 1:2, and nitrogen resource concentration was 0.4%, innoculation volume was 10%.Under this optimization conditions,the β-mannan mannohydrolase activity reached to 48.1IU/mL after fermentation for 4d, which was 2.3 times than the activity of control sample.
