摘要
研究了聚乙二醇(PEG-4000)/磷酸钾双水相体系中,PEG浓度、磷酸钾浓度和pH对双水相萃取法从黄粉虫中提取超氧化物歧化酶(SOD)的影响。采用改良的邻苯三酚自氧化法测定SOD活性,并用SDS-聚丙烯酰胺凝胶电泳测定SOD的分子量。结果表明:双水相体系的质量组成为PEG12%,磷酸钾24%,pH7.1时,SOD的萃取效果最好,SOD纯化倍数达3.74,活性回收率达71.2%。
The effects of concentration of polyethylene glycol, potassium phosphate and pH on extraction of superoxide dismutase (SOD) from the tenebrio molitor in aqueous two-phase system of PEG-4000 and potassium phosphate were studied. Superoxide dismutase activity was determined by improved pyrograllol autoxidation and the molecular weight of SOD was determined by SDS-polyacrylamide gel electrophoresis.The results showed that the optimum conditions were as follows: the mass composition two-phase aqueous system PEG 12%, potassium phosphate 24% ,pH7.1, SOD purification multiple could reach 3.74, the activity recovery rate could reach 7l.2%.
